Bismarck Brown Y powder dye is used in various staining methods in microscopy. It is a metachromatic dye used in histology for staining tissues, acid mucins, cartilage, and bones. It is also used in cytology. It is most commonly used as a counterstain to Gentian Violet.
The mentioned formulation is only one of the ways of preparing the dye solution. Bismarck Brown Y is most commonly used in exfoliative cytology and hematology. Depending on personal requests and standard laboratory operating procedures, the dye solution can be prepared according to other protocols.
Dissolve 0.5 g of Bismarck Brown Y dye in 80 ml of absolute ethanol Mix with 20 ml of hydrochloric acid, 1% aqueous solution .
